One was discharged but the other remained in critical condition, Devon and Cornwall police said.
The other two were checked and discharged by paramedics on the spot.
Police said they found and found the bodies during a “large-scale search” of Lake Roadford, near Okehampton.
Chief Superintendent Dan Evans of Devon and Cornwall Police said: “This is an extremely worrying incident and our thoughts are with everyone involved and their loved ones, with whom we are working closely to offer help and support where we can.
“Tragically, we can confirm that the bodies of two people were found and found this afternoon.
“Our priority is to support their families.
The page of the branch investigating maritime accidents on the government’s website gov.uk previously reported that two people with disabilities are unknown.
The families of the two, who were unaccounted for, were briefed on developments and supported by specially trained staff.
Investigators believe all of these people in the boat are locals in Devon.
